Can I start a custom bot from an ongoing conversation? If a user sends a reply to an open ongoing conversation, but his reply is off-hours, I need it to send a custom reply. How can I do that?

@carmel​ You cannot send/trigger custom bot in middle of a conversation. A probable option would be to create a separate bot and schedule it to be sent out of Office hours. This will be sent to all users who contact you during the time and who match the bot criteria. We have an article on how to set expectation for customers abut reply time and office hours.
